I got pregnant with my now 2-year-old while taking Prometrium to start AF for my first medicated cycle...but AF never came. No spotting, nothing. I did, however, spot in early pregnancy -- probably during weeks 6 to 8. Prometrium doesn't necessarily suppress ovulation, given that I definitely ovulated the first day or two that I was taking it, but it is progesterone --the dominant hormone during the luteal phase of the menstrual cycle (which occurs after ovulation) and during pregnancy (the placenta stimulates progesterone production during pregnancy).

Prometrium didn't cause my ovulation, but it didn't prevent it either. My gyn made sure that I understood that it provided no pregnancy protection when he first prescribed it for me to bring on AF. Hope that helps!
