VEGF gene variant increases risk for endometriosis

Investigating whether the +405 G/C polymorphism in the vascular endothelial growth factor gene is associated with endometriosis in the Italian population.


MedWIre News: Italian women are more likely to develop endometriosis if they carry the C allele of the +405 single nucleotide polymorphism (SNP) in the vascular endothelial growth factor (VEGF) gene, report investigators.

Previous findings on the association have shown conflicting results; one study in a South Indian population supported the association between the +405 G/C VEGF polymorphism and endometriosis susceptibility, whereas findings from a Korean study disproved the correlation.

To clarify the issue, Anna Maria Di Blasio (Istituto Auxologico Italiano, Milan, Italy) and team tested the distribution and frequency of three SNPs of VEGF-+405 G/C, +405 G/G and +405 C/C- in 203 women with advanced endometriosis and 140 healthy women.

A higher proportion of endometriosis sufferers carried the C allele of the +405 polymorphism compared with controls, at 40 versus 31 percent.

Based on a dominant model including women heterozygous and homozygous for the C allele, women carrying the C allele were almost twice as likely to develop endometriosis as women with the G/G genotype.

The presence of the C allele did not increase in line with severity of endometriosis, however.

"Considering the heterogeneity between the present and the two previous studies, these findings need to be further replicated in additional cohorts," the researchers report.
